I was messing around with the best buy demo, and i can't seem to figure it out. Is there even a way to distinguish between HOPO and regular notes?

The HOPO notes are a bit smaller than regular ones. It's hard to tell at first but they get a little easier.

They looked significantly smaller in the videos, but I haven't played the demo. Seems like a lot of people are having trouble with this?

After spending just a little time with the game, I find them to be as obvious as GH2, but not nearly as obvious as GH3.

They don't really need to be as obvious as they are in GH3 unless the placement's going to be as awkward as it was in GH3, so I guess that's coo.

they're not actually smaller than the other notes but the right and left edges have a white line along them reducing the amount of the note that is color, making it appear smaller

That's pretty cool, the GH3 HO/PO's are like blinding, covering the strum-required notes.

It takes probably an hour or so to get used to the HOPO notes in RB. They're about as obvious as in GH2.
